A Study on the Morphologic Reaction of the Trigeminal Ganglion After Experimental Surgery on the Maxillodental Region in Rats

Abstract
Trigeminal ganglia of normal rats and of adult rats subjected to total unilateral maxillary tooth extractions, a fracture of the anterior part of the mandible, and a fracture of the posterior part of the mandible were studied by light and electron microscopy.
Thirty two adult rats weighing about 200gm were used in this study. Animalss ire sacrificed 3, 14 and 30 days postoperatively by perfusion with 1% glutaraldehyde-paraformaldehyde in a phosphate buffer. The maxillary and mandibular parts of the trigeminal ganglia were removed and divided into small pieces which were rinsed in buffer, postfixed in osmium tetroxide, dehydrated in ethanol and embedded in Epon 812. One micron thick sections were cut from each block, stained with toluidine blue 0 and examined in the light microscope. Ultrathin sections were made from selected areas, stained with uranyl acetate and lead citrate, and examined in Hitachi H-500 electron microscope.
The results were as follows;
1. At 3 days after total unilateral maxillary tooth extractions, a small numbers of neurons showed sign of chromatolysis and pigment granules. At 14 days postoperatively,. a small numbers of neurons contained the nuclear membrane folding, swelling of nucleus and reduction of Nissl substance. At 30 days postoperatively, no evidence of ganglion cell degeneration was found.
2. At 3 days after anterior fracture of the mandible, a small numbers of cells interpreted as degenerating neurons displayed pigment granules and vacuoles. At 14 days postoperatively, a small numbers of neurons contained the shrinkage of the nucleus, hyperchromatosis of the nucleus, reduction of Nissl substance, and numerous lipofuscin granules. Most, of the affected neurons were small. During the 30 days of the anterior fracture of the mandible neurons appeared normal.
